It could be a tie tbh. But I think Isabella is slightly more physically able because of her Mama training. Then again Misato probably had some intense training when she first joined NERV. I'm going for Isabella because she's a total monster in the first few arcs of Promised Neverland.
Misato is just a human though. She doesn't use giant mechas or has any crazy abilities like regen or turning people into orange juice. She's literally on the very bottom tier of powerful evangelion characters

They should be relative in terms of physical condition. Misato kneed an experienced armored soldier to a wall, and Isabella can break limbs with little to no effort.
