Set one year after the events of Spider-Man 5, this film takes on a darker tone than the previous ones. It begins with a more aggressive and violent Peter Parker after Mary Jane's death, which caused him to suffer from Intermittent Explosive Disorder and Major Depression. He directly ends the lives of street thieves and criminals instead of bringing them to the police. Thirsty for revenge against supervillains, he takes out his fury on Shocker, a supervillain who caused social unrest in order to obtain Oscorp elements to enhance his suit with specialized combat gloves. Spider-Man defeats him in a battle, about to kill him. Shocker spares his life, reminding him that he is not the Spider-Man who murdered people, but rather brought them to justice. Shocker is taken to prison.
Later, there were rumors that Aleksei Sytsevich, a criminal mobster, traveled from Russia with interest in continuing his criminal life in New York, this criminal robs the bank with several henchmen and Spider-Man manages to stop the henchmen except for Sytsevich himself where he takes the opportunity to escape in a tunnel with a mocking tone, explosives where Spiderman could not enter. The police begin to question Spider-Man about his actions where J. Jonah Jameson takes advantage of worsening the image of Spider-Man, Parker on the verge of madness sees the need to give up being Spider-Man and remember him as an honest and good-hearted hero to the violent one he has now become, he begins to go to therapy and decides to start a life from scratch, where he begins to improve his relationship with Gwen Stacey lamenting the death of his loved ones, Harry Osborn, Mary Jane, in the past.
Later, Dr. Connors decides to test his experiments to regenerate his arm (research he conducted in Spider-Man 5). He suffers a severe attack, and by accident, the piece of the Symbiote that Dr. Connors had preserved is released and escapes through the sewers. He calls an ambulance, where it transforms into the Lizard, causing social chaos and destroying cars in the middle of the highways. The first battle against him begins, where Spider-Man is defeated by brute force and manages to stun the Lizard, who manages to escape through the sewers. Aleksei, upon discovering that his greatest threat, whom he could destroy with his plans, was Spider-Man, desires to reach his level in order to defeat him. He enters Oscorp with several thugs, where they surround Felicia Hardy and threaten to kill her if she doesn't help him obtain an advanced prototype that would make Spider-Man unable to defeat him. Felicia leads him to the prototype of the giant rhinoceros skin, discarded experiments that were never used again. Felicia warns him about his chemistry, and Alexei, ignoring her comments, proceeds to put it on. He feels the pain in his body and tries to remove it, but his skin is cloned with the Rhino's, all in one. Granting him superhuman strength and durability, he's ready to continue stealing more and taking over areas with his gangs to gain respect.
Spider-Man, tired of being a hero, blames himself for the responsibility of being one, everything he's been through, and the deaths he's witnessed. He reveals to Gwen that he was Spider-Man. Felicia arrives quietly and tells her about the attack he suffered. Felicia and Gwen motivate Peter to continue being a hero, rising once more with the last bit of faith. He has to face two colossal enemies who surpass him in strength and skill. On one side, The Lizard, who wants to turn them all into reptilians to live in a better world, and Rhino, with his robberies and leadership over criminal gangs. Complicated battles where the strongest win, but this time they must use cunning to win. Rhino, clumsily, is defeated, stuck in a heavy cement tank, with buildings exploding and falling on top of him. Gwen's father, who initially refused help, decides to help Peter. Gwen, Spider-Man, Felicia, and Captain Stacey with their police officers defeat the villains, subverting The Lizard. The Lizard manages to cut and kill Gwen's father.
Post-credits
Everyone, demotivated and sad, goes to the cemetery for Captain Stacey's grave. Spider-Man, already corrupted by previous losses, hugs a depressed Gwen. The symbiote continues through the sewers and goes down the drain where it reaches a kind of prison.

About 6 months after the events of Spider-Man 4, Felicia Hardy takes control of Osborn from her deceased father, Adrien Toomes/Vulture, and injects herself with some of the enhanced super-soldier serum she had taken from her father. Spider-Man, more experienced than before, continues to roam the streets with enthusiasm after having learned many things from the past and having matured emotionally. He continued fighting crime, catching thieves, and saving people from misfortune (one of them being Max Dillon). Saved by Spider-Man from being run over by a stolen car, Max, grateful, reveals to Spider-Man that he was a fan of his heroism and popularity, saying that he was at the tribute (an event that occurred in Spider-Man 3; the site of his kiss with Gwen Stacey). J. Jonah Jameson continued to portray Spider-Man as a self-serving criminal. Max was a typical charismatic boy with few friends. He worked for Oscorp as an electrician and, after an unexpected incident, suffered an electrical accident while on the job, turning him to ash and giving birth to Electro. He remained in a coma for a long time.
On the other hand, Felicia Hardy committed several robberies in different parts of the city with the aim of distracting Spider-Man and leading him into her trap. Spider-Man finds her, and Felicia seduces him into a deadly trap that would end his life. Spider-Man, using his spider-sense, detects what she was plotting, and they begin to battle. Spider-Man defeats her and ties her up with a web net. He removes Felicia's mask and, surprised, sees it was her. He asks her why she did it. She confessed that it was because she killed her father, even though she didn't have the need to steal; she wanted to get his attention. However, Spider-Man explains that it was an accident and that the Vulture had been trying to kill him all along. Felicia didn't accept the truth until much later. But from the center of the city, she sensed something was wrong, and that it was Electro arriving after waking up. Confused, she didn't know what was happening. Electro takes revenge on Spider-Man for misrepresenting him to others by labeling him a fraud. The battle begins, vowing to become a God that not even Spider-Man could defeat. At the end of the film, Electro kills Mary Jane. Only with the help of a reconciled Black Cat was he able to defeat Electro, having perhaps the toughest fight of all the films so far, against a formidable enemy with Advanced Electrokinesis. Despite Peter and Mary Jane dating twice more, Peter will have to face a greater loss (of the person he was supposed to marry).
Electro dies from electrical overload with water (fatal)
As the film progresses, Dr. Connors teaches Peter that Lizard DNA could regenerate limbs (by testing the injection in animals).
